I think this sentence tells us all what is happening on the Island.
My opinion is that Man in Black is God (or a God of Egypt mythology), who was imprisoned by Jacob on the Island. Why?
In the Bible, God wants to wipe out the humanity, and makes the flood happen (here is the water theme). But Humanity is saved anyway and God promises he will never do something like this again.
But what is God got mad at the humanity once more, and decided that this is it? He is furious about our lack of faith and the way we live, and want us all exterminated. Jacob, an angel, cannot let God do this and he finds a way to keep God on the island where his power is really limited and from which he cannot escape.
That would explain about everything on the show. Why Jacob acts the way he does, why Man in Black is so dangerous but not necessarily evil, and why all the religious themes and the dead talking to the living.
