Here goes:
The island represents a time bubble, a pinched off, self contained universe, originally created by a huge source of energy. It trapped a musician, a doctor, a pregnant woman, etc inside. Such bubbles form all the time and normally can't interact with the parent universe, but in this case there is a huge source of energy trapped within. People can be moved back and forth but the only ones that can survive for long are those that are near duplicates of the original trap-ees (trap-ists?). Jacob calls these duplicates "candidates".
MIB, whoever he is, has also been trapped on the island. He want's to get out into the Parent universe, something that is a violation (of physical laws? of time line cohesion?) and he has createda loophole: he has 1) cultivated one of the candidates (Locke) to match his persona as much as possible, 2) had that candidate killed, then 3) recreated himself as a duplicate. This will allow him to leave the island, provided he can recreate that original group around him.
But if he does leave the island, it will drastically mess with the parent time line and cause all kinds of disruption. He doesn't care though, he just wants off.
