I just finished the recent episode and after Locke had a conversation with Kate I had a thought, and it goes like this.
Jacob and Locke are brothers, they don't have to be but that's my thought either way. Jacob has always had his powers or he was given them by his mother. Locke killed his mother or was the cause of her death/going crazy.
Jacob offered Locke the chance to redeem himself by serving him/the island. Locke grew tired of Jacob but he could not leave or do anything about it because if he does not obey the rules (referenced by the little boy, who I believe to be a young Jacob) then he loses the gift which Jacob gave him, and thus dies or whatever he was given in return for his servitude(Dogen's sons life for example) will be taken away. He had to find the loophole because without using it he would either have not been able to kill Jacob or he would have lost his powers when he did.
The short of it, Jacob gave Locke the choice to redeem an action and in return he had to be his servant, if the rules are broken the powers are taken from them.
Another idea, which would be a bit stranger, would be if Locke killed his mom, Jacob resurected her as a trade for Locke's servitude, and now Widmore is bringing her back to the island. (Maybe Widmore also made a deal with Jacob which prevents him from going on the island, as he is still in the sub, which involved protecting their mother, but that's a bit more out there.)

The short of it, Jacob gave Locke the choice to redeem an action and in return he had to be his servant, if the rules are broken the powers are taken from them. I like this idea. It's kind of like...MIB was serving out his debt to Jacob, but then when he had Jacob killed he had to take back what he got from his side of the deal, so he lost his ability to change form as he did before and became stuck as Locke. Or something.
