I was gonna put this in theories, but as it is based on some spoilers I heard, I put it here, so people who don't want to hear spoilers won't see.

The numbers refer to points on a Becker-Hagens grid. A BH grid is an 'energy" map of the Earth http://www.refugeofthed**ned.com/earth-energy-grid.html It is not a scientific map, but many have used it to find connections between areas of great mystical value, asources of natural electromagnetism. Cuse has said "The island can be found at point 42 on the Becker-Hagens grid" which lies somewhere between Australia and the Western coast of Africa - they're not in the Pacific after all. popwatch.ew.com/popwatch/2006/04/lost_the_origin.html
But why the number sequence? Why six distinct numbers?
Well, the numbers possibly correspond to other places where the Dharma Innitiative may have tried to set up a research centre and failed; or provide a way to find the island. So, for example, if you begin at point 4, you follow the electromagnetic frequency to point 8, then to point 16, and so on, and you'll find the Island. This may be how Penny is attempting to find the Island, and the research station with the Brazilian portuguese guys might be at point 8
